SAH caproKit
The SAH caproKit™ is a complete system to investigate SAM / SAH binding proteins.
SAM is the major source of methyl groups in biological reactions and used by methyltransferases in many important physiological processes. For example, DNA/RNA and Histone methylation provide important mechanisms for the regulation of genetic activity, specific methylation sites mediate restriction enzyme resistance, the degradation of neurotransmitters as well as biosynthesis and degradation and de-toxification of many compounds is accompanied through the transfer of methylgroups.
S-adenosyl-L-homocysteine (SAH) has a similar Kd as SAM and is used as selectivity function instead of SAM, due to higher stability. The Capture Compound™ scaffold is attached to the N6 position of SAH, as many MTases tolerate modifications at this position (1).
The SAH caproKit™ includes the SAH Capture Compound, SAH competitor, all buffers, positive control protein, and streptavidin magnetic beads.
The Kit can be used to discover and profile SAM binding proteins from a variety of different sources. Downstream of the CCMS process, the proteins can be subjected to tryptic digests, MS or gel electrophoresis (2).
Please note, that CCMS reagents (caproKits™) will perform best when used with the optimized equipment (caproBox™ and caproMag™).
For the most reproducible results it is recommended to standardize parameters such as UV wavelength, distance between sample and light source, temperature and incubation time. This can best be accomplished using a caproBox™ for cross-linking.
